How do I make GLaDOS speak in my level (I tried playing one of her lines with ambient_generic but it didn't work).

That tutorial refers to soundscapes and music. You want to use 'scenes' or an ambient generic which points to the raw (note raw) file in the browser. **It will not** preview in Hammer but it should work in game. Refer to http://theportalwiki.com/wiki/GLaDOS_voice_lines to preview the lines online.
