Well one thing that i'd like to see in portal are exponential expansions of various ideas.

one example would be a better portal gun, that can now throw portals onto more types of surfaces.

I remember how satisfying it is in games like prince of persia to get an upgraded sword that now has the ability to cut right through walls.

so it makes sense to me to have a portal gun that has more features...

portals onto more surfaces... makes portals of differing sizes... gee if you make a small portal and a large one, do you come out the same size? or is that a way to get bigger and smaller? heh that would be hot if you could grow super big or tiny tiny... although it could be rather scary if you go into an infinite ceiling floor loop and keep shrinking for a few hours. hm. ok maybe not haha.

but to have an enhanced gun that shoots more than just two portals, or can shoot more than one set of two perhaps (ya that's better--more than one set of two) as well as a gun that can shoot portals onto new surfaces, and moving surfaces... ya. mm.

and the cake. they can make better cake.

Hmm, I like the idea of shrinking. I doubt very much they will give you more than one pair of portals, just because it will be so hard to track which ones are which (for the player)

Maybe something like gravity changing.

i think of the first portal as the embryonic stage of the story. if the enrichment center was the egg you hatch out of in the start of the epic story, then there's far more to come as we find out more about the aperture/black mesa universe.

About the portal shrinking, it looks like something right out of Mario bros. I don't really thinks it fits the portal atmosphere, plus, it can be a source of plenty of difficult situations to handle.
Players find portal glitches everyday, imagine how much glitches we would have with giants and ants characters!

Judging the end of "Portal 1", I think that probably "Portal 2" may feature rescue of others test subjects.

Chell could infiltrate other aperture science facilities to rescue her comrades. Maybe we would have a special portal you can place only at a "rescue point" that allow you to teleport your friends test subjects outside.

I personnally would HATE that (except if it is just "near the end" like the behind the scenes) because it goes against the "lab-rat" sensation that makes Portal Portal, but it is something that sounds possible.
